Universal prophylactic antitumor vaccination using stem cell membrane-coated nanoparticles

Summary

Most cancer vaccines are given after a tumor has been diagnosed, when immune suppression is already strong. This study tested whether a vaccine given beforehand could prevent tumors from growing. The researchers coated nanoparticles with membranes from induced pluripotent stem cells (iPSCs), which carry oncofetal antigens that are often present on many types of cancer but scarce in normal adult tissue. When loaded with an immune adjuvant and injected into mice, the particles activated dendritic cells, generated broad T-cell responses, and inhibited tumor growth across five different mouse cancer models and in a bilateral model with two different tumors. The results suggest that a single stem-cell-membrane-based vaccine can provide broad prophylactic protection in mice, supporting further exploration of this approach for cancer prevention.
