Inhalable mRNA nanoparticles
Nature Materials2023Vol. 22278-279
Summary
This News & Views article discusses a study by Rotolo et al. that screened a library of 166 poly(β-amino esters) to identify an inhalable polymer nanoparticle formulation (P76) for delivering mRNA to the lungs. The formulation was effective and safe in multiple animal species, including mice, hamsters, ferrets, cows, and monkeys. In a hamster model of SARS-CoV-2 infection, treatment with P76 nanocomplexes loaded with CRISPR-based RNA protected against weight loss and outperformed a control antibody treatment. The findings highlight the potential of inhalable mRNA nanoparticles for treating lung diseases.
